چکیده

Background: This work aimed at determining whether bisphosphonate therapy produces a sufficient condition for jaw osteonecrosis after tooth extraction. Methods: Rats were allocated into 3 groups: (1)11 rats treated with alendronate; (2)10 rats treated with zoledronic acid; (3)10 control rats. The animals were submitted to tooth extractions, and at the end of bisphosphonate therapy, they were euthanized. Histological sections of the surgical site were processed and analyzed. Results: The zoledronic acid group showed higher incidences of osteonecrosis, inflammatory infiltrate and microorganisms. There was no significant difference for epithelial or connective tissue, root fragments, vital bone and positive staining for vascular endotheial growth factor (VEGF) among the groups. Conclusions: Zoledronic acid is associated with jaw osteonecrosis, whereas alendronate did not produce a condition sufficient for osteonecrosis after tooth extraction. Neither zoledronic acid nor alendronate was associated with a reduced immunohistochemical expression of VEGF in vital bone at the tooth extraction site.

Microscopic Evaluation of the Effect of Oral Microbiota on the Development of Bisphosphonate-Related Osteonecrosis of the Jaws in Rats

OBJECTIVES Osteonecrosis of the jaws is a side effect associated with the use of bisphosphonates. Using histologic analysis, this study aimed to evaluate the influence of microbial colonies in the development of osteonecrosis in the jaws of rats subjected to nitrogenous and non-nitrogenous bisphosphonates, undergoing surgical procedures. MATERIAL AND METHODS Thirty-four rats (Rattus norvegicu...

Avascular mandibular osteonecrosis in association with bisphosphonate therapy: a report on four patients.

UNLABELLED Over the past three years, several reports have been published on jaw osteonecrosis possibly being associated with the administration of bisphosphonates. Bisphosphonates are highly active inhibitors of osteoclasts. These drugs are used for the treatment of multiple myeloma, bone resorption in the case of metastatic malignant diseases, tumor-associated hypercalcaemia, and in the treat...

Pharmacogenetics of bisphosphonate-associated osteonecrosis of the jaw.

An undesirable effect associated with bisphosphonates is osteonecrosis of the jaw (ONJ). Case reports discussed ONJ development in patients with multiple myeloma or metastatic cancers receiving bisphosphonates as palliation for malignant bone disease. No causative relationship has been unequivocally demonstrated between ONJ and bisphosphonate therapy. To determine if a higher sensitivity to bis...

Bisphosphonates and Osteonecrosis of the Jaw.

BACKGROUND Since 2003, reports have been published on necrosis of the jaw bones possibly being associated with the administration of bisphosphonates. Bisphosphonates are highly active inhibitors of osteoclasts which have been used prophylactically or symptomatically in the treatment of plasmocytoma, bone metastasis of malignant disease, tumor-associated hypercalcaemia and in the treatment of os...

Denosumab-related Osteonecrosis of the Jaws: a Case Report

Bisphosphonates are potent and effective drugs frequently used to prevent the skeletal complications associated with postmenopausal osteoporosis in women, to manage patients with multiple myeloma, hypercalcemia, and metastasis of cancer to the bone. The search for new medications with the same therapeutic effectiveness as the bisphosphonates but fewer side effects has resulted in the discovery ...
